Each species of archaea and bacteria possesses a 16S gene, responsible for encoding a tiny subunit of ribosomes. This gene contains regions (V1 to V9) that show significant variability and are used for taxonomic categorization purposes. The gene also has conserved portions that can act as targets for primers to amplify highly variable regions. The 16S rRNA approach is now widely used to analyze complex microbial communities thanks to the implementation of next generation sequencing (NGS). Next generation sequencing (NGS) is the most effective method to study any microbiome due to the limitations of Sanger sequencing and culture-based methods. Sanger sequencing cannot detect many bacteria in a single sample, and culture-based methods are time-consuming and often cannot be cultured for certain types of bacteria. The EasySeq™ 16S Microbiome Library Preparation Kit uses reverse complement PCR (RC-PCR) technology, offering a cost-effective and efficient methodology for characterizing the microbiome. This kit allows you to analyze the V3-V4 hypervariable regions in one reaction, all in one tube. The main rationale for using the V3-V4 regions is their high nucleotide variability and excellent discriminatory power, which are essential for assessing bacterial diversity and quantifying microbial abundance in different sample types.
