Reverse complement polymerase chain reaction (RC-PCR) is a high-throughput library preparation method used to analyze hypervariable regions, also known as “mutation hotspots,” in extremely small amounts of nuclear mitochondrial DNA (mtDNA) that is highly degraded. This method is especially useful for complex materials that do not have a complete short tandem repeat (STR) profile. For laboratories in countries where rules or regulations prohibit whole genome sequencing for forensic purposes.
